Carolme 20 AM tablets contains Olmesarton Medoxomil and Amlodipine. Olmesarton Medoxomil mainly reduces hypertension and antagonize the action of angiotensin II receptor. Amlodipine is a calcium channel blocker which helps in dilation of the blood vessels which causes increase in the blood flow.
The drug can be taken with or without food with the starting dose of 5-20 mg once daily.
Mechanism of action
It produce its action by inhibiting the binding of angiotensin II to AT I. This causes AT I mediated vasoconstriction which causes decrease in the blood resistance and blood pressure.
It mainly produces its action by inhibiting the influx of calcium ions by calcium channel which causes arterial smooth muscle contractibility and causes vasoconstriction. This causes vasodilation which decrease in blood pressure.
The drug after oral administration is bioactivated by ester hydrolysis to olmesarton and which causes absorption from gastrointestinal tract.
The volume of distribution for the drug is about 17 ltrs. The drug is mainly bound to plasma protein and causes its penetration in red blood cells.
The drug is mainly excreted through urine and feces.
After oral administration it slowly absorbed from GI tract. The peak plasma concentration is attained in 6-12 hrs. Co- administration of the food doesn't effect the action of the drug. The bioavailability of the drug is about 64-90%.
The drug produces its action by binding to the protein. The protein binding of the drug is about 97.5 %. The drug follow hepatic enzymes for its metabolism.
The drug metabolites are excreted mainly through urine.
